Basic Operation Guide of HITACHI H-7650 Transmission Electron Microscope

I. Prechecks and specimen loading

1) Check the POWER status of the system: make sure the POWER key switch is at COL. ON position.

2) Check the vacuum EVACs are solid green.

3) Check the Main Hitachi TEM Control window for any error messages. Report to center staff for any trouble shooting/ error.

4) Remove specimen holder from the goniometer: toggle the EVAC/AIR switch from EVAC to AIR. You will see the red light on followed by “hiss” sound. Slowly pull holder straight out.

5) Load specimen holder to the goinmeter:

i. Insert the specimen holder into the goniometer with the guide pin aligned correctly. After pushing the specimen holder to its first stop, toggle the EVAC/AIR switch to EVAC.

ii. Wait until the EVAC green light is on (it takes about 1-2 min), then turn specimen holder clockwise until it stops. The specimen will be pulled in by the vacuum.

iii. Leave the specimen holder in this position until the beam is turned on and aligned.

II. Applying beam

1) From the menu bar, click Operate-HV control-ACC. Voltage to open HV control window, click Operate-HV control-Filament to open filament control window. Click function-PDS to bring out alignment settings for different kV.

2) Choose desired voltage file in PDS and then click on load. The HV should start ramping up. When it is done, click on the filament current ON in the filament control window. You should see beam on the florescence screen at this point.

3) Insert your specimen holder under the beam by turning specimen holder counterclockwise, then it will be pulled in.

4) Press wobble button, use the pin under goniometer to bring the sample height to the Eucentric point.

III. Imaging

Navigate and find specimen image on the florescence screen first. Switch to the channel 2 computer, open the AMT software for camera imaging. Click on “Camera OUT” on the upper right corner to flip the florescence screen and start imaging by clicking “click for live image”.

To capture image, click on the “click for final image”, and then save it under File-save as.

Click on the capture final image again to go back to live imaging.

IV. Sample exchange

Turn the filament off. Pull the specimen holder out gently until it stops, then turn clockwise until it stops. Pull it further out then counter clockwise. Wait until the light turn from red to green. Then switch it to AIR to take out the holder.

V. Shutting down

Click on “Camera in” to bring back the florescence screen. Switch to channel 1 computer. Filament off, HV off.

VI. Data transfer

You may bring in a flash drive to copy data from the channel 2 computer.
